簡介
外泌體(Exosome)是來源于膜的納米級(jí)囊泡,包含了復(fù)雜 RNA 和蛋白質(zhì)的小膜泡,直徑在30至150nm。它們是在正常及病理?xiàng)l件下由多種類型的細(xì)胞釋放的,其主要來源于細(xì)胞內(nèi)溶酶體微粒內(nèi)陷形成的多囊泡體,經(jīng)多囊泡體外膜與細(xì)胞膜融合后釋放到胞外基質(zhì)中。外泌體對(duì)細(xì)胞功能的影響已經(jīng)與各種生理過程相關(guān)聯(lián),包括:細(xì)胞間通訊、癌癥轉(zhuǎn)移、免疫調(diào)節(jié)活性以及傳染物(如朊病毒和逆轉(zhuǎn)錄病毒)的傳播。
外泌體可從多種體液中回收,如血漿、血清、尿液、精液、唾液、支氣管液、腦脊液、乳汁、羊水、滑膜液、淚液、淋巴液、膽汁和胃酸等。外泌體可在相鄰和遠(yuǎn)端細(xì)胞之間傳遞蛋白質(zhì)和遺傳信息。外泌體可穿過細(xì)胞膜、不易引起免疫反應(yīng)特點(diǎn),使其在基因、抗腫瘤等藥物的體內(nèi)運(yùn)送方面具有獨(dú)特優(yōu)勢。
外泌體組分不僅包括脂質(zhì)和蛋白質(zhì),還包括核酸(主要是miRNA和mRNA),也有一些報(bào)道發(fā)現(xiàn)外泌體當(dāng)中存在基因組和線粒體DNA。外泌體可以將信號(hào)傳遞到受體細(xì)胞,從而介導(dǎo)一種新的細(xì)胞間通訊機(jī)制。外泌體在各種生物過程中發(fā)揮著重要作用,因此,對(duì)于外泌體深入研究與轉(zhuǎn)化應(yīng)用,外泌體的分離富集與表征鑒定是重要一步。

外泌體表征鑒定相關(guān)抗體產(chǎn)品
鑒定外泌體首先需要通過WB來鑒定外泌體的標(biāo)志蛋白是否存在于樣品中,通過電子顯微鏡來觀察樣品中細(xì)胞外囊泡的形態(tài)特征以及直徑,對(duì)其進(jìn)行區(qū)分鑒定。
